基于HPLC-MS/MS 技术测定不同品种枸杞中黄酮类物质的研究

摘要:通过优化液相和质谱条件,并考察不同的提取溶剂、提取方式和称样量,建立了枸杞中6种黄酮类物质的液相色谱-质谱检测方法。6种黄酮类物质在各自标准曲线浓度范围内线性良好,相关系数(r)在0.9911~1.0000之间。枸杞鲜果中6种黄酮类物质的平均回收率为72.5%~118.4%,相对标准偏差(RSD)为0.7%~14.2%;枸杞干果中6种黄酮类物质的平均回收率为71.7%~109.0%,RSD为0.9%~11.3%。考察了10个枸杞品种中6种黄酮类物质的含量,结果差异明显。该试验为进一步研究枸杞黄酮类物质的活性及加工产品的开发提供数据支撑,为枸杞产业高质量发展提供科学依据。

Study on Determination of the Flavonoids in Different Varieties of Lycium barbarum Based on HPLC-MS/MS Technology

Abstract: A liquid chromatography-mass spectrometry method for the determination of 6 flavonoids in Lycium barbarum was established by optimizing the conditions of liquid phase and mass spectrometry, and different extraction solvents, extraction method and sample weight were investigated. 6 flavonoids showed good linearity within their respective standard curve concentration ranges,the correlation coefficients (r) ranged from 0.991 1~1.000 0. In fresh Lycium barbarum, the average recovery rates of 6 flavonoids were 72.5%~118.4% and the relative standard deviations (RSD) were 0.7%~14.2%. In dried Lycium barbarum,the average recovery rates of 6 flavonoids were 71.7%~109.0% and the RSD were 0.9%~11.3%. The content of 6 kinds of flavonoids in 10 varieties of Lycium barbarum was investigated,the result showed great difference. This experiment provided data support for further research on the activity of Lycium barbarum flavonoids and the development of processed products,and scientific basis for the high quality development of Lycium barbarum industry.
