常见预制肉制品肉源掺假可视化LAMP方法的建立

摘要:为实现猪、牛、羊、马、鸡、鸭6种常见预制肉制品肉源掺假的现场快速检验,基于环介导等温扩增技术(LAMP),分别选取6种高保守序列为靶标,设计特异性引物,建立并优化LAMP反应体系,通过引入酚红实现扩增结果可视化,并验证方法的特异性、灵敏度、检出限和稳定性。将可视化LAMP方法与实时荧光PCR法同时用于市售预制肉制品的掺假检验,对比分析检测结果。结果表明,该研究建立的可视化LAMP方法特异性强、稳定性好,其中牛、羊、猪的基因检测灵敏度为1fg/μL,马、鸡的检测灵敏度为0.1fg/μL,鸭的检测灵敏度为0.01fg/μL,且均能检出0.1%的掺假。在市售预制肉制品的掺假比对检验中,该研究方法与国家标准实时荧光PCR法的结果一致性达100%。该研究建立的可视化LAMP方法可高效、准确、便利地鉴别预制肉制品中的6种掺假成分,为基层肉源掺假的现场快速检验提供新方法。

Establishment of the LAMP Method for Visualizing Meat Source Adulteration in Common Premade Meat Products

Abstract: To achieve rapid on- site test of meat source adulteration in 6 common premade meat products,namely, pig, cow, sheep, horse, chicken and duck, based on the loop- mediated isothermal amplification(LAMP) technology, 6 kinds of highly conserved sequences were selected as targets, specific primers were designed,the LAMP reaction system was set up and optimized,and the visualization of the amplification results was achieved through the introduction of phenol red, and the method’s specificity, sensitivity, limit of detection,and stability were verified. The visualized LAMP method and the real time fluorescence PCR method were simultaneously used for the adulteration test of commercially available premade meat products,and the test results were compared and analyzed. The results showed that the visualized LAMP method established in this study had high specificity and good stability,in which the sensitivity of gene detection was 1 fg/μL for cow,sheep and pig,0.1 fg/μL for horse and chicken,and 0.01 fg/μL for duck,and all of them could be detected with 0.1% adulteration. In the adulteration comparison test of commercially available premade meat products,the results of this study method were 100% consistent with the national standard real time fluorescent PCR method. The visualized LAMP method established in this study can efficiently,accurately and conveniently identify the 6 adulterated components in premade meat products, and provide a new method for rapid on- site test of adulteration of meat sources at the grassroots level.
