雁荡山铁皮石斛多糖结构分析及其对斑马鱼胃肠黏膜炎症和损伤的修复作用研究

摘要:以雁荡山1号铁皮石斛为原料,通过提取纯化得到铁皮石斛多糖,采用紫外光谱、傅里叶红外光谱、凝胶色谱和离子色谱对其结构进行分析;采用斑马鱼模型探究铁皮石斛多糖对胃肠的作用。结果表明,经提取纯化得到一种纯度较高的铁皮石斛多糖,命名为DOP1,其纯度为95.0%,分子量(Mw)为3.882kDa。其单糖组成比例为阿拉伯糖0.40%、半乳糖0.42%、葡萄糖39.03%、甘露糖60.11%。通过斑马鱼模型实验证明了铁皮石斛多糖对胃肠黏膜损伤具有一定的修复作用,具体表现为斑马鱼的胃肠道面积减小,中性粒细胞减少和杯状细胞增加。

Structural Analysis of Dendrobium officinale Polysaccharides from Yandangshan and Study on Its Repairing Effect on Inflammation and Damage of Zebrafish Gastrointestinal Mucosa

Abstract: Dendrobium officinale polysaccharides were obtained by extraction and purification by using Dendrobium officinale Yandangshan No.1 as raw material. Its structure was analyzed by UV,FTIR,gel chromatography and ion chromatography; The zebrafish model was used to investigate its effects on the gastrointestinal tract. The results showed that a high purity Dendrobium officinale polysaccharide named DOP1 was obtained from extraction and purification, the purity was 95.0%, and the molecular weight (Mw) was 3.882 kDa. The monosaccharide composition ratio was 0.40% of arabinose, 0.42% of galactose, 39.03% of glucose, and 60.11% of mannose. Through zebrafish model experiments, it has been proven that Dendrobium officinale polysaccharides have a certain repairing effect on gastrointestinal mucosal damage. It is specifically showed in reduction of gastrointestinal area,neutropenia and increasing goblet cells in zebrafish.
