摘要:建立一种保健食品中黄芪甲苷、毛蕊异黄酮葡萄糖苷、甜菜碱、补骨脂素、异补骨脂素和淫羊藿苷的检测方法。样品经甲醇40 ℃超声提取60 min,采用水(A)和0.1%甲酸乙腈(B)作为流动相,利用超高效液相色谱-三重四极杆质谱联用仪,选用电喷雾离子(electron spray ionization,ESI+)源、正离子检测模式(positive mode,MRM)进行测定。结果表明,6种功能成分的回收率为85.7%~108.5%,该方法实验室内变异系数为0.91%~4.32%(n=7),方法检出限为0.02~0.60 μg/kg。该方法准确、灵敏、高效,适合于中药类保健食品中特征成分黄芪甲苷、毛蕊异黄酮葡萄糖苷、甜菜碱、补骨脂素、异补骨脂素和淫羊藿苷的分析检测。

Determination of Six Functional Components in Health Food by Ultra-high Performance Liquid Chromatography-triple Quadrupole Mass Spectrometry

Abstract: To establish a method for the detection of astragaloside IV, calycosin-7-O-β-D-glucoside, betaine, psoralen, isopsoralen and icariin in health food. The samples were ultrasonically extracted with methanol at 40 ℃ for 60 min, water(A) and 0.1% formic acid acetonitrile(B) were used as mobile phases. The samples were determined on ultra performance liquid chromatography triple quadrupole mass spectrometer by electron spray ionization(ESI+)source and positive ion detection mode (positive mode,MRM). The results showed that the recoveries of the six functional components were 85.7%~108.5%, the intra-laboratory coefficients of variation were 0.91% ~4.32% (n=7), and the detection limits were 0.02~0.60 μg/kg, respectively. The method is accurate, sensitive and efficient, and it is suitable for the analysis and detection of the characteristic components astragaloside IV, calycosin-7-O-β-D-glucoside, betaine, psoralen, isopsoralen and icariin in traditional Chinese medicine health food.
