黄连生物碱复合物及其单体通过调节肠道菌群改善小鼠肥胖病症的研究

摘要:为了比较黄连素、黄连碱两种单体及黄连生物碱复合物对肥胖小鼠体重的影响及肠道菌群的变化,本研究将 50 只小鼠随机分为空白组(C)、模型组(M)、黄连素处理组(Ber)、黄连碱处理组(Con)和黄连生物碱处理组(Trc),每组10只,除空白组外,其余4组给予高脂饮食,建立实验室肥胖小鼠模型。造模14 d后,每天给予140 mg/L的药物干预,空白组及模型组给予等剂量的生理盐水。随机选取3只观测指标,并分别于0 d7 d14 d21 d28 d称取小鼠体重,测量血清中TCTG含量的变化,同时检测小鼠体内肠道菌群的变化。研究结果表明:空白组和模型组小鼠的重量在第14 d时有明显差异,造模成功;实验第28 d,黄连素、黄连碱和黄连生物碱处理的小鼠重量与模型组相比有显著差异,黄连素、黄连碱和黄连生物碱处理能有效改善肥胖小鼠的体重及血清中TCTG的含量,调节血脂水平,且黄连生物碱处理组相比黄连素和黄连碱处理组,指标变化明显,说明黄连生物碱药物干预更有助于改善小鼠肥胖;M组小鼠肠道内Bacteroides属、Lactobacillus属、Akkermansia属、Desulfovibrio属及Prevotella1属相比C组有显著差异,药物干预后,呈显著上升趋势,且Trc组效果更明显。

The Research on Coptis Alkaloid Complex and its Monomers Improve Obesity in Mice by Regulating Gut Microbes

AbstractIn order to compare the effect of berberine, coptisine and berberine alkaloid complexes on weight of obese mice and changes in intestinal flora, 50 mice were randomly divided into the control group (C), the model group (M), the berberine treatment group (Ber), the coptisine treatment group (Con) and the berberine alkaloid treatment group (Trc), with 10 mice in each group. In addition to the control group, the other 4 groups were given high-fat diet to establish an experimental obese mouse model. After 14 days for modeling, 140 mg/L of drug intervention was given to the groups with berberine, coptisine and berberine alkaloids treatment group, and isodose saline was given to the control group and the model group. Randomly selected three mice for observation indicators, the mass of body was weighted at 0 d, 7 d, 14 d, 21 d and 28 d, and the changes of TC and TG contents in the serum were measured. The changes of intestinal flora in the mice were also detected. The results showed that the body mass of mice in the control group and the model group was significantly different on the 14th day, which indicated successful molding. On the 28th day of the experiment, the weight of the mice treated with berberine, coptisine and berberine alkaloids was significant different compared with the model group. The berberine, coptisine and berberine alkaloids can effectively improve the weight of obese mice, the content of TC and TG in serum, and regulate blood lipid levels, and the indicates changes more obviously by berberine alkaloids treated compared to the berberine and coptisine treated. The Bacteroides, LactobacillusAkkermansia, Desulfovibrio and Prevotella in the intestine of group M mice have significant different compared to group C. After drug intervention, the indicators significant upward, and the effect of the Trc group is more obvious.
