产碱性纤维素酶菌株WS-135的筛选及酶学性质研究

摘要:对实验室保藏菌种短小芽孢杆菌WS-135进行紫外诱变,筛选出产酶高的菌株A-5,酶活力为22.1 U·mL-1,较未诱变菌株提高2.37倍。以紫外诱变筛选菌株为出发菌株,进行硫酸二乙酯诱变,筛选得到一株酶活为28.9U·mL-1的菌株B-7,较未诱变菌株提高1.3倍。B-7菌株连续传代8次,酶活力保持稳定。研究其酶学特性,结果表明温度为35 ℃时酶的活性最好,在pH7-10时,有较好的酶活性;在40 ℃pH 9.010.0条件下酶活稳定,在常温下保藏8个月后,仍有80%的酶活力,表明该酶稳定性较好。

Screening of Alkaline Cellulase Producing Strain WS-135 and Study on Its Enzymatic Characterization

AbstractThe original strain Bacillus pumilus WS-135 was mutated by UV in the paper, screening of high enzyme producing strain A-5, and its enzyme productivity was 22.1 U·mL- 1 and 2.37 fold higher than that of parent strain. The mutant A-5 was mutated by DES, and a strain B-7 with 28.9 U·mL- 1 enzyme activity was obtained which was 1.3 fold higher than that of parent strain. The activity of alkaline cellulase remained stable after 8 passages. The optimal temperature and pH of alkaline cellulase were at 35 ℃, pH 7.0-10.0. The activity of alkaline cellulase was stable at 40 ℃, pH 9.0 and pH 10.0. Store for 8 months at 25 ℃, and enzyme activity of alkaline cellulase reduced 20% when compared with the initial enzyme activity.
