摘要:该文对延龄草总皂苷的最佳提取方法进行筛选,并通过紫外分光光度法与高效液相色谱法对发酵前后药液进行对比分析,在单因素实验的基础上,采用Box-Behnken响应面实验研究了黑曲霉接种量、发酵时间和发酵温度对延龄草中总皂苷提取率的影响。结果表明:延龄草总皂苷最佳提取方法为75%乙醇回流法,其最佳发酵条件为黑曲霉接种量2.25%,发酵时间6d,发酵温度27 ℃,在此条件下总皂苷提取率为8.620%,与预测结果基本一致。

Study on Fermentation Technology and Extraction Conditions of Total Saponins from Trillium tschonoskii Maxim. by Aspergillus niger

Abstract:Firstly,screen the optimal extraction method for total saponins from Trillium tschonoskii Maxim. Then by using UV and HPLC to compare and analyze the fermentation solution before and after fermentation,Box-Behnken response surface analysis was used to study the effects of Aspergillus niger inoculation amount, fermentation time, and fermentation temperature on the extraction rate of total saponins in Trillium tschonoskii Maxim., which based on a single factor experiment. The results showed that: The best extraction method for total saponins in Trillium officinale is 75% ethanol reflux method,and the optimal fermentation conditions for total saponins of Trillium tschonoskii Maxim. were Aspergillus niger inoculant amount of 2.25%,fermentation time of 6 d and fermentation temperature of 27 ℃. Under these conditions,the extraction yield was 8.620%,which was basically consistent with the predicted results.
