摘要:探究绿豆大曲的活性成分及其对急性醉酒小鼠乙醇代谢和肝脏的具体影响与机制。通过超高压液相色谱串联四级杆飞行时间质谱(UPLC-Q-TOF-MS)技术分析绿豆大曲中潜在活性成分;建立急性醉酒小鼠模型,采用行为学实验、酶联免疫吸附实验(Enzyme linked immunosorbent assay, ELISA)、实时荧光定量聚合酶链反应(Real-time quantitative polymerase chain reaction, RT-qPCR)、自动生化分析技术探究绿豆大曲及活性成分对小鼠乙醇代谢和肝脏的具体影响;采用网络药理学技术结合蛋白免疫印迹实验(Western blot, WB)进一步探究绿豆大曲影响急性酒精性肝病的可能机制。结果显示,绿豆大曲存在32种潜在活性成分,具有促进小鼠乙醇代谢和醒酒的作用潜力,可减轻小鼠饮酒带来的肝脏负担,例如氧化应激、血脂紊乱、转氨酶升高及炎症因子分泌增多,这可能与其中活性成分协同抑制PI3K/AKT/MAPK信号通路相关。

The Study on the Effect and Mechanism of Lvdou Daqu Active Components on Ethanol Metabolism and Liver Injury in Acute Intoxicated Mice

Abstract: To investigate the active components of Lvdou Daqu and their specific effects and mechanisms on ethanol metabolism and liver in acute intoxicated mice. The potential active components in Lvdou Daqu were analyzed by Ultra-Performance Liquid Chromatography-Quadrupole Time-of-Flight Mass Spectrometry (UPLC-QTOF-MS); a mouse model of acute intoxication was established, and behavioral experiments, enzyme linked immunosorbent assay (ELISA), real-time quantitative polymerase chain reaction (RT-qPCR), and automated biochemical analysis techniques were used to study the specific effects of Lvdou Daqu on ethanol metabolism and the liver. The possible mechanisms of Lvdou Daqu affecting acute alcoholic liver disease were further investigated using network pharmacology technique combined with western blot assay (WB) . The results showed the presence of 32 potential active components in Lvdou Daqu, and they had the potential effect to promote mice’s metabolism of ethanol and sobriety and could reduce the burden of the liver caused by alcohol consumption, such as oxidative stress, dyslipidemia, elevated aminotransferases, and increased secretion of inflammatory factors, that may be related to the synergistic inhibition of PI3K/AKT/MAPK signaling pathway from active components.
