摘要:采用水提醇沉、树脂纯化的方法提取纯化党参多糖,并对其进行多糖含量、分子量分布及单糖组成的检测分析,以及基于斑马鱼巨噬细胞模型的免疫调节功能研究。结果表明:党参多糖含量为88.63%,其纯度较高、均一性好,重均分子量为1.138×104;主要由6种单糖组成,包括:D-无水葡萄糖、D-甘露糖、D-半乳糖醛酸、半乳糖、L-(+)-阿拉伯糖、鼠李糖,其中D-无水葡萄糖占比最大,占单糖总量的53.28%;党参多糖在100μg/mL时具有促进斑马鱼巨噬细胞吞噬作用的功效,低于50μg/mL的党参多糖对斑马鱼巨噬细胞吞噬作用影响较小。

Study on the Extraction and Purification of Codonopsis pilosula Polysaccharides and Its Immunomodulatory Activity

Abstract: The polysaccharide of Codonopsis pilosula was extracted and purified by water extraction, ethanol precipitation and resin purification,the polysaccharide content,molecular weight distribution and monosaccharide composition were detected and analyzed, and the immunomodulatory function was studied based on zebrafish macrophage model. The results showed:the content of Codonopsis pilosula polysaccharide was 88.63%,with high purity and good uniformity, and weight average molecular weight was 1.138×104. It is mainly composed of 6 monosaccharides, including D- anhydrous glucose, D- mannose, D- galacturonic acid, galactose, L-(+)-arabinose and rhamnose, among them, D- anhydrous glucose accounted the most for 53.28% of the total amount,when Codonopsis pilosula polysaccharide was 100 μg/mL,it had the effect of promoting the phagocytosis of zebrafish macrophages, Codonopsis pilosula polysaccharides less than 50 μg/mL had lower effect on the phagocytosis of zebrafish macrophages.
