摘要:为了提高农用木霉的生产效率,该研究建立了哈茨木霉H7的固体发酵罐生产工艺。结果表明,主原料为小麦(浸泡12h),辅料为1.25%甘油、1.75%蛋白胨和0.005%硫酸锰;最适发酵条件为:发酵温度30℃、接种量10%、初始pH 5;在此条件下,在150L固体发酵罐中发酵后得到的产孢量为39.0×108 CFU/g。通过番茄的萌发和盆栽实验结果表明,浓度为0.01×108 CFU/mL孢子液提升了番茄种子的萌发活力;浓度为0.1×108 CFU/mL孢子液促进了番茄幼苗生长,鲜重、茎粗、株高、根长、总叶绿素含量均有显著性提高(P<0.01)。该研究为哈茨木霉H7的规模化生产和应用提供了参考。

Optimization of Solid-state Fermentation Process of Trichoderma harzianum H7 and Study on Its Growth-promoting Effect on Tomatoes

Abstract: To enhance the production efficiency of agricultural purpose Trichoderma, this study established a production process of solid fermentation tank for Trichoderma harzianum H7. The results indicated that the optimal medium composition consisted of wheat (soaked for 12 h) as the main substrate, supplemented with 1.25% glycerol, 1.75% peptone, and 0.005% MnSO4. The optimal fermentation conditions were: temperature 30 ℃,inoculum amount 10%,and initial pH 5. Under these conditions,a sporulation yield of 39.0×108 CFU/g was achieved in a 150 L solid fermentation tank. Tomato germination and pot experiments demonstrated that a spore suspension concentration at 0.01×108 CFU/mL enhanced seed germination vigor,while its concentration at 0.1×108 CFU/mL promoted seedling growth,with substantial increases (P<0.01) in fresh weight,stem diameter, plant height,root length,and total chlorophyll content. This study provides a reference for the mass production and application of Trichoderma harzianum H7.
