响应面法对极端东方化假单胞菌产嗜铁素发酵条件的优化

探索极端东方化假单胞菌SWNY 3-6产嗜铁素的发酵条件,旨在为嗜铁素的深入应用研究及其在农业生物防治领域的推广应用提供数据支撑。本研究以嗜铁素相对含量为评价指标,在培养温度、发酵时间、pH、通气量单因素实验基础上,利用Box-Behnken中心组合原则,设计四因素三水平响应面实验,对其发酵条件进行优化。结果表明,优化后的发酵条件为培养温度35.5℃、发酵时间38.5 hpH7.2、通气量1.8 vvm,在此条件下嗜铁素相对含量达到了59.36%,接近模型预估值。通过响应面法优化极端东方化假单胞菌产嗜铁素的发酵条件,确定产嗜铁素的最优发酵条件,为后续生产及应用研究提供理论依据。

Optimization of the Fermentation Condition for Siderophores Production 

by Pseudomonas extremorientalis by Response Surface Methodology

Abstract: In this study, we explored the fermentation conditions of Pseudomonas extremorientalis SWNY3-6 for siderophores production, aiming to provide data support for the further application of siderophores and its popularization and application in the agricultural biological control field. Relative content of siderophores was applied as the evaluation index. Firstly, the separate experiments of culture temperature, fermentation time, pH and ventilation was conducted, respectively. Then the response surface methodology was designed with four factors and three levels to optimize the fermentation conditions, using the Box- Behnken central combination principle. The obtained optimal fermentation conditions were 38.5 h fermentation at 35.5 , with a pH value of 7.2 and ventilation 1.8 vvm. Under these conditions, the relative content of siderophores reached 59.36%, close to the predicted value of the model. The optimal fermentation conditions for siderophores production by Pseudomonas extremorientalis were determined, which provided a theoretical basis for subsequent production and application research.
