大蒜素通过GSK-3β/β-catenin信号通路抑制舌鳞状细胞癌EMT的研究

:本研究旨在探讨大蒜素(Allicin)对舌鳞状细胞癌上皮向间充质转化(EMT)的影响,并探究其作用机制。本实验采用CCK-8法检测不同剂量大蒜素对HSC-3HSC-4细胞的生存作用并计算半数抑制作用(IC50);划痕和Transwell小室实验测定大蒜素对细胞的迁移能力和侵袭影响;Western Blot检测蛋白表达。结果显示,大蒜素对HSC-3HSC-4细胞活性有抑制作用且具有时间和剂量依赖性;不同浓度(204060 μmol/L)的大蒜素能够抑制HSC-3HSC-4细胞迁移和侵袭;Western Blot结果显示大蒜素能够升高EMT标志物 E-cadherin表达,降低N-cadherinVimentin表达,同时对GSK-3β/β-catenin信号通路中P-GSK-3ββ-catenin蛋白起到抑制作用。结果表明,大蒜素能够抑制舌鳞状细胞癌HSC-3HSC-4细胞侵袭和迁移,其机制可能是通过抑制GSK-3β/β-catenin信号通路逆转EMT发生实现的。

Study of Allicin Inhibiting EMT of Tongue Squamous Cell Carcinoma Through the GSK-3β/β-catenin Signaling Pathway

Abstract: This study aims to investigate the effect of allicin on epithelial-mesenchymal transition (EMT) of tongue squamous cell carcinoma and explore its mechanism. HSC-3 and HSC-4 cells were treated with different concentrations of allicin. CCK-8 assay was used to detect the effect of allicin on the survival of HSC-3 and HSC-4 cells, as the same time, the half-maximal inhibitory concentration ( IC50 ) was calculated. The effect of allicin on cell migration and invasion was detected by wound healing and Transwell assay. Western Blot was used to detect the protein expression. The results showed that allicin inhibited the viability of HSC-3 and HSC-4 cells in a time and dose-dependent manner. Allicin at different concentrations ( 20, 40, 60 μmol/L ) could inhibit the migration and invasion of HSC-3 and HSC-4 cells. Furthermore, Western Blot result showed that allicin increased the expression of E-cadherin, decreased the expression of N-cadherin and Vimentin, and inhibited the protein expression of P-GSK-3β and β-catenin in the GSK-3β/β-catenin signaling pathway. Allicin could inhibit the invasion and migration of tongue squamous cell carcinoma HSC-3 and HSC-4 cells, and the mechanism may be achieved by reversing EMT by inhibiting GSK-3β/β-catenin signaling pathway.
