基于酿酒废水发酵生产棘孢木霉工艺条件优化及应用研究

摘要:在单因素实验的基础上,基于响应面法对酿酒废水(底锅水、黄水)发酵生产棘孢木霉工艺条件进行优化研究,确定基于酿酒废水生产棘孢木霉最佳工艺条件:pH5.3,接种量为8.0%,发酵时间为3.8 d。在此条件下测得棘孢木霉生物量为6.76×107 CFU/mL,与响应面分析模型预测值(6.52×107 CFU/mL)接近。优化结果对于酿酒废水资源化利用生产棘孢木霉菌剂具有实际参考价值。进一步对酿酒废水发酵生产棘孢木霉污泥减量应用效果评价,对过程污泥浓度与胞外聚合物变化进行分析,并通过扫描电镜观察,结果表明发酵生产的棘孢木霉对活性污泥有较好的破解减量效果。

Optimization of Fermentation Conditions for Trichoderma echinospora Production from Liquor-making Wastewater and Its Application

AbstractOn the basis of single factor experiment, based on the response surface optimization experiment, the technological conditions of producing Trichoderma echinospora by fermentation of liquor-making wastewater (steamer bottom water、Huangshui) were optimized. The optimum technological parameters of producing Trichoderma echinospora based on liquor-making wastewater were determined: pH 5.3, inoculation amount 8.0%, fermentation time 3.8 days. Under these conditions, the biomass of Trichoderma echinospora was 6.76×107 CFU/mL, which was close to the predicted value of 6.52×107 CFU/mL in the response surface analysis model. The optimization results had practical reference value for the utilization of liquor-making wastewater to produce Trichoderma echinospora. Further, through the evaluation of the application of Trichoderma echinospora produced by fermentation of liquor-making wastewater during the sludge reduction test was carried out, the change of the sludge concentration and the extracellular polymer was analyzed, and the scanning electron microscope observation showed that Trichoderma echinospora had a good effect on the reduction of activated sludge.
