摘要:建立超高效液相一测多评法(UPLC-QAMS)同时测定淡豆豉配方颗粒中大豆苷、黄豆黄苷、染料木苷、大豆苷元、黄豆黄素、染料木素的含量,验证其准确性及适用性。采用UPLC法,色谱柱为Waters CORTECS UPLC T3(2.1 mm×100 mm,1.6 μm);流动相为乙腈-0.1%甲酸,梯度洗脱;流速为0.3 mL/min;柱温为30 ℃;检测波长为260 nm。以染料木苷为内参物,建立该成分与其他5个成分的相对校正因子,利用相对校正因子计算各成分的含量,同时对一测多评法的计算值和外标法的实测值进行比较。结果表明,相对校正因子的耐用性良好,一测多评法与外标法测得11批样品中异黄酮成分的含量结果无显著性差异。所建立的一测多评法简便稳定,结果准确,适用于淡豆豉配方颗粒的定量分析及质量控制。

Simultaneous Determination of Six Isoflavones in Dandouchi Formula Granules by Quantitative Analysis of Multi-Components with Single-Marker

Abstract:To establish an ultra-high performance liquid chromatography-quantitative analysis of multi-components by single marker (UPLC-QAMS) method for simultaneous determination of daidzin, glycitin, genistin, daidzein, glycitein, genistein in Dandouchi formula granules, and to verify its accuracy and feasibility. UPLC method was used, the determination was performed on CORTECS UPLC T3 (2.1 mm×100 mm, 1.6 μm) column with mobile phase consisted of acetonitrile-0.1% formic acid solution, gradient elution; the volume flow rate was 0.3 mL/min, the column temperature was 30 ℃; the detection wavelength was 260 nm. Genistin was used as an internal standard in the UPLC method, and the relative correction factors of the other five components were established, which were adopted to calculate the mass fraction of each. The results of QAMS method and the external standard were compared. The results indicate that the reproducibility of the relative correction factors was good, and there was no significant difference between the QAMS method and external standard in determining the contents of isoflavones in 11 batches of samples. The established UPLC- QAMS method is simple, stable and accurate, which can be employed for quantitative analysis and quality control of Dandouchi formula granules.
