摘要:优化调味品等的前处理方法,建立可同时快速测定 9 种甜菊糖苷类甜味剂的高效液相色谱(high performance liquid chromatography,HPLC)分析方法。样品经乙腈-水(1∶1,v/v)溶解,超声辅助提取,乙酸锌-亚铁氰化钾盐析,采用Shim-pack C18色谱柱分离,乙腈-0.01 mol/L磷酸二氢钠缓冲液(pH=2.6)(32∶68)洗脱,紫外测定,外标法定量。结果表明,9种甜菊糖苷类甜味剂在20.0~400.0 mg/L范围内,线性关系良好,相关系数R2均大于0.996,加标回收率在73.3%~101.2%之间,相对标准偏差(relative standard deviation,RSD)为2.1%~5.3%,方法检出限、定量限分别为5.0~15.0 mg/kg、15.0~45.0 mg/kg。该方法前处理简单快速,重复性好,精密度高,适用于调味料等中多种甜菊糖苷类化合物的测定,填补国内多种甜菊糖苷类化合物检测的空白,为食品安全监管及风险评估提供技术支撑。

Simultaneous and Rapid Determination of Nine Stevia Glycosides Compounds in Condiments by HPLC

Abstract:To establish a method for the simultaneous and rapid determination of nine stevioside sweeteners in condiments by high performance liquid chromatography (HPLC) using optimized sample pretreatment. Samples were dissolved by acetonitrile-water (1∶1, v/v), and extracted by ultrasound, after salted-out by zinc acetate-potassium ferricyanide, the separation of nine stevia glycosides was performed on a Shim-pack C18 column with acetonitrile-0.01 mol/L sodium dihydrogen phosphate buffer (pH=2.6) (32∶68) and quantified by external standard method with ultraviolet. A good linear relationship was observed in the range from 20.0 to 400.0 mg/L, the correlation coefficient R2 was greater than 0.996, the recoveries of spiked standard were from 73.3% to 101.2%,the relative standard deviation (RSD) were from 2.1% to 5.3%, the limit of detection and the limit of quantity were from 5.0 to 15.0 mg/kg and from 15.0 to 45.0 mg/kg. The method has the advantages of simple, rapid, reproducible and precise, which can suit for the determination of stevia glycosides in condiments, fill the blank of detection of various stevia glycosides in China, and provide technical support for food safety supervision and risk assessment.
