发酵铁皮石斛花醇提物的抗氧化和美白活性研究

摘要:探究微生物发酵对铁皮石斛花功效活性的影响。分别将毕赤酵母、酿酒酵母、乳酸菌三株菌对铁皮石斛花进行纯种发酵,以80%乙醇为溶剂进行超声提取得到发酵铁皮石斛花乙醇提取物。测定其黄酮和总酚含量,通过DPPHABTS自由基清除能力和总抗氧化能力测定,探究其发酵前后的抗氧化活性和抑制酪氨酸酶活性的美白功效。结果表明,经毕赤酵母发酵后的铁皮石斛花乙醇提取物中黄酮含量最高为13.17 mg/g,经酿酒酵母发酵铁皮石斛花乙醇提取物中总酚含量最高为1.04 mg/g。随着提取物浓度增大,提取物整体抗氧化能力逐渐增强,在1 mg/mL条件下,酿酒酵母发酵铁皮石斛花乙醇提取物的DPPH自由基清除率最高达81.5%,对ABTS+自由基清除率最高为66.9%,总抗氧化能力最强为1.03 mmol/g,乳酸菌发酵铁皮石斛花乙醇提取物的酪氨酸酶抑制率最高为37.8%

Study on Antioxidant and Whitening Activities of Alcohol Extracts from Fermented Dendrobium candidum Flowers

AbstractThe aim was to explore the effect of microbial fermentation on the functional activity of dendrobium officinale flowers. Pichia farinosa, Saccharomyces cerevisiae and Lactococcus lactis were respectively used for performing pure culture fermentation on the Dendrobium officinale flowersthe ethanol extract of fermented Dendrobium candidum flowers was obtained by ultrasonic extraction with 80% ethanol as the solvent. The contents of flavonoids and total phenols were determined. The DPPH free radical, ABTS free radical scavenging capacity and total antioxidant capacity were measured to explore the antioxidant activity before and after fermentation and the whitening effect of inhibiting tyrosinase activity. The highest flavonoid content in the ethanol extract of Dendrobium candidum flowers fermented by Pichia pastoris was 13.17 mg/g, and the highest total phenol content in the ethanol extract of Dendrobium candidum flowers fermented by Saccharomyces cerevisiae was 1.04 mg/g. With the increase of extract concentration, the overall antioxidant capacity of the extract was gradually enhanced. Under the condition of 1 mg/mL, the DPPH free radical scavenging rate of ethanol extract from Dendrobium candidum flowers fermented by S. cerevisiae was 81.5%, the highest clearance rate of ABTS free radical was 66.9% , the strongest total antioxidant capacity was 1.03 mmol/g, and the highest tyrosinase inhibition rate of ethanol extract of Dendrobium candidum flowers fermented by lactic acid bacteria was 37.8%.
