HPLC法同时测定辣木叶提取物中8种酚酸类成分含量

摘要:建立HPLC法同时测定辣木叶提取物中新绿原酸、绿原酸、隐绿原酸、维采宁-2、牡荆素、异牡荆素、异槲皮苷、紫云英苷的含量,为辣木叶提取物的质量评价提供参考。以80%甲醇作为提取溶剂,超声提取40 min。采用Waters Atlantis T3 色谱柱(4.6 mm×250 mm5 μm),甲醇-0.1%甲酸作为流动相梯度洗脱,流速 1.0 mL/min,柱温30 ,检测波长300 nm;进样量5 μL。结果8种酚酸类化合物成分在各自范围内线性关系良好(r0.999 9),平均加标回收率95.64%99.16%RSD值在0.95%3.35%之间。实验结果表明该方法操作简单,准确度高,重复性好,可用于辣木叶提取物样品中的含量测定及产品质量监控。

Simultaneous Determination of 8 Phenolic Acids in Moringa oleifera Leaf Extract by HPLC

AbstractTo established an HPLC method to simultaneously determine the contents of neochlorogenic acid, chlorogenic acid, cryptochlorogenic acid, vetenin-2, vitexin, isovitexin, isoquercitrin, and astragalin in extracts of Moringa oleifera leaves. To provide a reference for the quality evaluation of Moringa oleifera leaf extract. Using 80% methanol as extraction solvent, ultrasonic extraction for 40 min. And then performed on a 30  thermostatic Waters Atlantis T3 column (4.6 mm×250 mm, 5 μm), with the mobile phase comprising of methanol and 0.1% Formic acid flowing at 1.0 mL/min in a gradient elution manner, and the detection wavelength was set at 300 nm. A volume of 5 μL upper solution was injected into HPLC for analyzing. The 8 kinds of phenolic acid compounds have a good linear relationship within their respective ranges (r0.999 9), the average recovery rate of addition is 95.64%99.16%, and the RSD value is between 0.95%3.35%. The experimental results show that the method is simple to operate, accurate, and reproducible, and can be used for the content determination of Moringa oleifera leafextract samples and product quality monitoring.
