摘要:建立HPLC法同时测定赶黄草药材及醇提物中没食子酸、儿茶素、槲皮苷、槲皮素的含量,为赶黄草药材及醇提物的质量评价提供参考。以60%甲醇作为提取溶剂,超声提取30 min。采用YMC色谱柱(4.6 mm×250 mm,5 μm),乙腈-0.1%磷酸作为流动相梯度洗脱,流速1.0 mL/min,柱温30 ℃,检测波长205 nm;进样量10 μL。结果表明,4 种化合物成分在各自范围内线性关系良好(r≥0.999 9),平均加标回收率 96.15%~100.21%,RSD 值在0.74%~1.79%之间。实验结果表明该方法操作简单,准确度高,重复性好,可用于赶黄草药材及醇提物样品中的含量测定及产品质量监控。

Simultaneous Determination of the Contents of 4 Active Components in Penthorum chinense and Its Alcohol Extracts by HPLC

Abstract:To establish an HPLC method to simultaneously determine the contents of gallic acid, catechin,quercetin, and quercetin in the materials and alcohol extracts of Penthorum chinense, and provide reference for the quality evaluation of materials and alcohol extracts of Penthorum chinense. With 60% methanol as the extraction solvent, ultrasonic extraction was performed for 30 min. Use YMC column (4.6 mm×250 mm, 5 μm) Acetonitrile 0.1% phosphoric acid was used as the mobile phase for gradient elution, the flow rate was 1.0 mL/min, the column temperature was 30 ℃, and the detection wavelength was 205 nm; Injection volume 10 μL. The four compounds have a good linear relationship within their respective ranges (r≥0.999 9), the average recovery rate of standard addition is 96.15%~100.21%, and the RSD value is between 0.74%~1.79%. The experimental results show that the method is simple to operate, accurate, and reproducible. It can be used for the content determination and product quality monitoring of Penthorum chinense materials and alcohol extract samples.
