摘要:以大豆卵磷脂与胆固醇为原料,通过薄膜分散法制备包载AHLL的脂质体。以AHLL脂质体的包封率作为考察指标,通过单因素实验对AHLL脂质体的制备配方进行选择,根据实验综合结果选择的较优配方为脂质体卵胆比为5∶1,药脂比为1∶8,超声时间5 min,AHLL 药物总浓度为1 mg/mL。在此基础上制备得到的脂质体粒径为(115.88±6.35)nm,包封率为(81.16±1.31)%,电位为-24.1 mV。透射电镜(TEM)观察脂质体呈分散均匀的圆球形,初步稳定性实验表明脂质体在4 ℃下具有较好的贮藏稳定性,在胃肠液中可以有效保护AHLL的生理活性,并且被脂质体包覆的AHLL具有良好的缓释效果。

Preparation and Stability Evaluation of Liposomes for Encapsulation of Active Peptides

Abstract:The liposomes encapsulated with AHLL were prepared by a thin-film dispersion method using soy lecithin and cholesterol as raw materials. The encapsulation rate of AHLL liposomes was used as the index of investigation, and the formulation of AHLL liposome preparation was selected by single-factor experiment, and the better formulation was selected based on the comprehensive results of the experiment, which was 5∶1 liposomal oocyte ratio, 1∶8 drug-lipid ratio, 5 min sonication time, and 1 mg/mL total drug concentration of AHLL. The average particle size of the liposomes prepared on this basis was (115.88±6.35) nm, the encapsulation rate was (81.16 ± 1.31)%, and the potential was - 24.1 mV. Transmission electron microscopy (TEM) showed that the liposomes were uniformly dispersed in a circular shape. Preliminary stability experiments showed that the liposomes had good storage stability at 4 ℃, could effectively protect the physiological activity of AHLL in gastrointestinal fluid, and the AHLL encapsulated by liposomes had good slow release effect.
