响应面法优化红曲菌液态发酵产莫纳可林K工艺

摘要:以紫色红曲菌为出发菌株,在200 L发酵罐中进行液态深层发酵生产莫纳可林K(简称MK)。为得到含量最高的MK,在发酵培养基不变的条件下,分别以罐压、通气量和搅拌转速为因素进行单因素试验,确定因素水平后,以MK为评价指标进行Box-Behnken响应面优化,确定该菌株产MK的最佳中试发酵工艺:罐压为0.06 MPa,通气量为22 m3/h,搅拌转速为134 r/min。结果表明,在此条件下,发酵液中MK含量为828 mg/L。本研究获得红曲菌液态深层发酵产MK的最佳中试发酵工艺,为大生产及后续研究提供理论依据。

Optimization of Monacolin K Production by Liquid Fermentation of Monascus Using Response Surface Methodology

AbstractMonascus purpureus was used as the starting strain to produce Monacolin K (MK) by liquid fermentation in 200 L fermenter. In order to obtain the highest MK, the single-factor experiment was carried out with the tank pressure, the aeration rate and the agitation speed as the factors under the condition of constant fermentation medium, the optimization of Box-Behnken response surface was carried out with MK as the evaluation index, and the optimal medium-scale fermentation process was determined as follows: tank pressure 0.06 MPa, aeration 22 m3/h, agitation speed 134 r/min. The results showed that the content of MK in the fermentation broth was 828 mg/L. In this study, the optimum medium- scale fermentation process of Monascus MK production by liquid fermentation was obtained, which provided theoretical basis for large-scale production and follow-up research.
