摘要:利用高通量测序技术对患根腐病三七及健康三七根际土与根内生真菌群落组成,物种丰富度及多样性分析。根际土中测序共获得204818高质量序列,主要分为子囊菌门,被孢霉门,球囊菌门、担子菌门及毛霉门;健康三七根际土中被孢霉门,球囊菌门和担子菌门相比健康三七均有着较高的丰度,而子囊菌门则有着较低的丰度。根内共获得209079有效序列,主要分为子囊菌门、担子菌门和毛霉门,根内生真菌的多样性低于根际土的真菌多样性。且患病三七根内致病菌镰刀菌属丰度显著低于根际土中镰刀菌属丰度,可能是镰刀菌属为土壤致病菌导致的。另外,发现无论是在根际土还是根内,患病三七中球囊菌门丰度均高于健康植株,因此,对球囊菌门更深一步的研究有助于探讨球囊菌门与三七根腐病的关系。

Illumina High-throughput Sequencing Reveals Fungal Community Composition and Diversity in Root and Rhizosphere Soil of Health and Root-rot Panax notoginseng

Abstract:Using the Illumina High-throughput sequencing technology to reveal fungal community composition and diversity in root and rhizosphere soil of health and root-rot Panax notoginseng. A total of 204818 effective tags were obtained from rhizosphere soil, and the community was composed of five phyla (Ascomycota, Mortierella, Glomus, Basidiomycota and Mucor); The relative abundance of Mortierella, Glomus and Basidiomycota in health Panax notoginseng was higher than root-rot Panax notoginseng, and the Ascomycota was lower than root-rot Pannx notoginseng. A total of 2049079 effective tags were obtained from root, which were mainly divided into Ascomycota, Basidiomycota and Mucor. The diversity of endophytic fungi in root was lower than the diversity of rhizosphere soil. Moreover, in the root-rot Panax notoginseng, the abundance of Fusarium in the root was significantly lower than in rhizosphere soil, it may be caused by the soil pathogen Fusarium. In addition, we found that whether in the root or in the rhizosphere soil, the abundance of Glomerella in the root-rot Panax notoginseng was higher than the healthy Panax notoginseng. Therefore, the research on the Glomerella is helpful to explore the relationship of Glomerlla and root-rot in Panax notoginseng.
