天麻醇提物对灰树花深层培养发酵中胞外多糖的影响

摘要通过在灰树花液态深层发酵体系中添加冻干天麻醇提物,测定发酵过程中胞外多糖、菌丝体生物量、还原糖含量和pH值变化,分析天麻中四种主要成分天麻素、对羟基苯甲醇、对羟基苯甲醛和巴利森苷发酵过程中的变化。研究发现,添加9 g/L冻干天麻醇提物的灰树花发酵液比空白组(未添加天麻醇提物)发酵液菌丝体生物量和胞外多糖含量分别显著增加了29.43%31.49%P0.05);经分析,9 g/L的冻干天麻参与灰树花发酵属于生长部分偶联型,且在动态发酵的第7 d时,灰树花胞外多糖和菌丝体开始大量积累,分别达到了1.565±0.035 g/L1.370±0.041 g/L;发酵12 d后天麻四种成分天麻素、对羟基苯甲醇、对羟基苯甲醛以及巴利森苷含量显著降低,推测天麻四种成分基本被灰树花生长代谢所利用。结果表明,冻干天麻醇提物可以促进灰树花胞外多糖和菌丝体生物量的合成,为提高灰树花高值利用提供一定的参考。

Effect of Ethanol Extract of Rhizoma gastrodiae on Exopolysaccharide in Submerged Fermentation of Grifola frondosa

AbstractTo explore the influence of Rhizoma gastrodiae alcohol extract on the metabolism of Grifola frondosa submerged fermentation. Add the freeze-dried alcohol of Rhizoma gastrodiae extract to the liquid submerged fermentation system of Grifola frondosa, to explore the dynamic changes of exopolysaccharide, biomass, reducing sugar content and pH value, and analyze the participation of Rhizoma gastrodiae in the fermentation process the changes in the content of its four main components gastrodin, p-hydroxybenzyl alcohol, p-hydroxybenzaldehyde and parishin. The study showed that the mycelial biomass of Grifola frondosa fermentation broth with 9 g/L freeze-dried alcohol extract of Rhizoma gastrodiae was significantly increased by 29.43% and the content of exopolysaccharide was significantly increased by 31.49%that compared with the blank group (no addition of Rhizoma gastrodiae) (P0.05). On the 7th day of dynamic fermentation, (1.565±0.035) g/L of Grifola frondosa extracellular polysaccharides and (1.370±0.041) g/L of biomass began to accumulate in large quantities, 9 g/L freeze- dried Rhizoma gastrodiae participating in the fermentation of Grifola frondosa belongs to the growth part coupling type; on the 12th day of fermentation, the contents of gastrodin, p-hydroxybenzyl alcohol, p-hydroxybenzaldehyde and parishin are significantly reduced. It is speculated that the four components of Rhizoma gastrodiae are basically absorbed and utilized by the growth and metabolism of Grifola frondosa. The results showed that the components of freeze-dried Rhizoma gastrodiae can be used to promote Grifola frondosa the synthesis of exopolysaccharide and biomass, which can provide a certain reference for improving the high-value utilization of Grifola frondosa.
